Title DENERVATION IN MURINE FAST-TWITCH MUSCLE: SHORT TERM PHYSIOLOGICAL CHANGES AND TEMPORAL EXPRESSION PROFILING Authors

چکیده
Denervation deeply affects muscle structure and function, the alterations being different in slow and fast muscles. Since the effects of denervation on fast muscles are still controversial and high throughput studies on gene expression in denervated muscles are lacking, we studied gene expression during atrophy progression following denervation in mouse tibialis anterior (TA). The sciatic nerve was cut close to trochanter in adult CD1 mice. 1, 3, 7 and 14 days after denervation animals were sacrificed and TA muscles were dissected out and utilized for physiological experiments and gene expression studies. Target cDNAs from TA muscles were hybridized on a dedicated cDNA microarray of muscle genes. Seventy-one genes were found differentially expressed. Microarray results were validated and the expression of relevant genes not probed on our array was monitored by RQ-PCR. Nuclearand mitochondrialencoded genes implicated in energy metabolism were consistently down-regulated. Among genes implicated in muscle contraction (myofibrillar and sarcoplasmic reticulum) genes typical of fast fibers were down-regulated, whereas those typical of slow fibers were up-regulated. Electrophoresis and Western blot showed less pronounced changes in myofibrillar protein expression, partially confirming changes in gene expression. Isometric tension of skinned fibers was little affected by denervation, whereas calcium sensitivity decreased. Functional studies in mouse EDL muscle showed prolongation in twitch time parameters and shift to the left in force-frequency curves after denervation. We conclude that, if studied at mRNA level, fast muscles appear not less responsive than slow muscles to the interruption of neural stimulation.

متن کاملHyperpolarized Functional Magnetic Resonance of Murine Skeletal Muscle Enabled by Multiple Tracer-Paradigm Synchronizations
Measuring metabolism's time- and space-dependent responses upon stimulation lies at the core of functional magnetic resonance imaging. While focusing on water's sole resonance, further insight could arise from monitoring the temporal responses arising from the metabolites themselves, in what is known as functional magnetic resonance spectroscopy. Performing these measurements in real time, howe...
متن کاملChronic low-frequency stimulation upregulates uncoupling protein-3 in transforming rat fast-twitch skeletal muscle.
The purpose of this investigation was to examine the temporal changes in uncoupling protein (UCP)-3 expression, as well as related adaptive changes in mitochondrial density and fast-to-slow fiber type transitions during chronically enhanced contractile activity. We examined the effects of 1-42 days of chronic low-frequency electrical stimulation (CLFS), applied to rat tibialis anterior (TA) for...
متن کاملFunctional reorganization of the partially denervated hindlimb extensor and flexor muscle in rat.
In view of the neuromuscular system plasticity the functional changes induced by partial denervation are presented. The long-term effects of partial denervation of postural (soleus-SOL) or flexor (extensor digitorum longus-EDL) muscles on their EMG activity were studied in rats. The activity per motor unit was significantly higher both in the partially denervated SOL and EDL muscles. During sta...
متن کاملTemporal Correlation Of Bax Expression And Axotomy-Induced Motoneuronal Apoptosis In Adult Rats: A Morphometric, Ultrastructural And Immunohistochemical Study
Background and Objective: As apoptotic cell death is extremely involved in physiological development and many pathological situations such as cancer and neurodegenerative diseases, the understanding of its molecular machinery can be useful in designing new therapeutic strategies. The present study investigated the temporal expression of the proapoptotic protein Bax in adult spinal motoneuron...
